Although Synchronize Folder still isn't enabled on the internal drive.
Reverting to a backup of the Lightroom catalog seemed to fix it.

Weird: All of a sudden Lightroom won’t auto import, auto import dialog raises exception, drag import doesn’t work, Sync Folder disabled.
